Differential release of sialic acid from normal and malignant cells by Vibrio cholerae neuraminidase or influenza virus neuraminidase.

چکیده
A comparison was made between the amounts of sialic acid released from viable cell suspensions of normal and malignant cells by Vibrio cholerae neuraminidase (VCN) and by influenza virus neuraminidase. More sialic acid can be removed by enzyme digestion from various tumors than from normal lymphoid cells. More sialic acid can be removed from the surface of both normal and tumor cells by VCN than by influenza virus neuraminidase. VCN lyses the 2-3, 2-6, and 2-8 glycosidic linkages between sialic acid and the mucopolysaccharides of the cell surface, and influenza virus neuramini dase lyses only the 2-3 and 2-8 linkages. Therefore, it appears that most cell-surface sialic acid residues are bound by the 2-6 linkage on both normal and malignant cells. The relative increase in cell-surface sialic acid residues may be important to tumor immunotherapy, since the removal of cell-surface sialic acid residues with VCN increases the immunogenicity of tumor cells.

Neuraminidase gene sequence analysis of avian influenza H9N2 viruses isolated from Iran
Influenza A viruses possesses two virion surface glycoproteins including haemagglutinin (HA) and neuraminidase (NA). The NA plays an important role in viral replication and promotes virus release from infected cells and facilitates virus spread throughout the body. To find out any genomic changes that might be occurred on NA gene of avian influenza circulating viruses, we have genetically analy...
متن کاملAction of Vibrio cholerae neuraminidase (sialidase) upon the surface of intact cells and their isolated sialolipid components.
The effect of strong electrolyte upon the activity of Vibrio cholerae neuraminidase (VC neuraminidase) toward the surface of intact cells and toward their isolated, purified sialolipid components, was studied in detail. A linear dependence of the logarithm of initial velocity on the square root of ionic strength was observed for purified GM8 over a concentration range of 0 to 0.05 M NaCl in 0.0...
متن کاملSerum Levels of Sialic Acid and Neuraminidase Activity in Cardiovascular, Diabetic and Diabetic Retinopathy Patients
Background: Sialic acid is a component of serum that is elevated in diseases such as diabetes and certain malignancies. The normal range of SSA concentration and serum neuraminidase activity in different populations are varied, probably due to racial differences. Objective: The purpose of the present study was to obtain the average SSA concentration and serum neuraminidase activity, in an Iran...

Surface sialylation and glycosylation of tumor cells is known to affect various biological phenomena. In the present study, we analyzed the regulatory roles of cell surface sialylation in cell adhesion to galectin-1 in the human diffuse large B cell lymphoma (DLBCL) cell line, HBL-2, and Burkitt's lymphoma cell line, HBL-8. Vibrio cholerae neuraminidase treatment enhanced HBL-2 cell adhesion to...
متن کاملMembrane sialic acid and the mechanism of insulin action in adipose tissue cells. Effects of digestion with neuraminidase.
Mild digestion of isolated adipose tissue cells with Closfridium perjringens neuraminidase (10 to 20 mpg of purified enzyme per ml, 15 min at 37”) results in enhanced glucose transport by these cells. This effect appears to mimic closely the physiological effect of insulin on glucose transport by normal cells, and there is evidence to suggest that these two apparently different stimuli may affe...
